Assessment of antimicrobial, anti-diabetic, anti-Alzheimer and anti-cancer properties, along with bioaccessibility and bioactive potential of Polygonum cognatum Meissn extracts from various cities in Türkiye

This study aimed to evaluate the total phenolic content (TPC), total condensed tannins (TCT), and antioxidant activities as well as in vitro bioaccessibility, antimicrobial, anti-diabetic, anti-Alzheimer, and cytotoxic properties of madımak crude extracts collected from different provinces of Türkiye. The results showed that the crude extract of TPC values were 25.03–40.07 mg GAE/g, with bioaccessibility index (BI) values of 17.14%–29.12%. The TCT values varied from 20.09 to 61.91 mg CAE/g, accompanied by BI values ranging from 7.41% to 22.50%. For ABTS antioxidant activity, the values spanned from 113.70 to 339.40 mg TE/g, with BI values between 8.48% and 27.18%. The FRAP antioxidant values were recorded between 149.15 and 348.87 mg TE/g, with BI values ranging from 9.54% to 19.86%. The concentration of macroelements (mg/100 g) was highest for potassium (410.67–503.00), followed by calcium (201.67–307.00), magnesium (56.67–88.67), and sodium (1.87–4.13). For microelements (mg/100 g), the order of concentration was iron (25.07–44.10), phosphorus (22.33–47.67), zinc (0.53–0.94), and copper (0.34–0.67). Madımak exhibited antibacterial efficacy against Bacillus cereus (0.312–2.5 mg/mL), Staphylococcus aureus (0.039–1.156 mg/mL), Escherichia coli (0.312–2.5 mg/mL), Salmonella Typhimurium (0.156–1.25 mg/mL), and Listeria monocytogenes (0.126–2.5 mg/mL). The madımak samples demonstrated impressive inhibitory activity against α-glucosidase (51.54–66.56%) and α-amylase (69.94–84.63%). Additionally, these samples exhibited low cytotoxic effects on HEK-293 cells, with IC50 values ranging from 1595.54 to 3096.42 μg/mL. Overall, these findings highlight madımak's potential as a valuable natural source with diverse health benefits and promising health-related properties, making them viable for industrial utilization as food additives.
