N-acetyl cysteine and metal nanoparticles internalization: A critical methodological aspect

Metal nanoparticles have toxic effects on different cell types. Antioxidants exert ameliorative effect on the toxicity of metal nanoparticles. N-acetyl cysteine has antioxidant and metal ion-chelator properties and reduce nanoparticle toxicity. This study aims to address this ambiguity, whether N-acetyl cysteine pretreatment exert ameliorative effect on metal nanoparticles toxicity via antioxidant properties and/or inhibition of nanoparticle internalization. Human umbilical vein endothelial cells were cultured and pretreated with N-acetyl cysteine (2, 10, 50 mM) for 4 h and then, cells were treated with TiO 2 , CeO 2 , and CuO nanoparticles (100 μg/ml). After 24 h incubation, intracellular ions concentration were measured by inductively coupled plasma atomic emission spectroscopy (ICP-OES) and considered as nanoparticles internalization. Our results showed that N-acetyl cysteine at the higher concentration (50 mM) enhance nanoparticle internalization, while lower concentration of N-acetyl cysteine (2 and 10 mM) have no significant effect on nanoparticle internalization. Therefore, N-acetyl cysteine in pretreatment model at the concentration lower than 10 mM is applicable for MNPs toxicology.
