Özet

Prolonged and significant reduction of renal blood flow may cause acute renal failure (ARF). There is a common consensus that different pathogenetic mechanisms are responsible from the structural and functional abnormalities in ischemic ARF. The aim of this study was to evaluate the effects of ketanserin on renal functions in ischemic ARF by using vascular clamp technique. This study was performed on male wistar rats (n=20, BW: 290±12 g) chosen according to their body weights. This study was performed in four periods. 1. Basal period: (time 60 min, drug : 0.9% NaCl); 2. Occlusion period : (time 60 min, no drug); 3. Infusion period : (time 60 min, Group-I: Control , n=10 , drug : 0.9 % NaCl ; Group-II : Ketanserin, n=10 , drug : ketanserin 0.05 mg/kg IV bolus and 0.1 mg/kg/h IV infusion ; 4. Recovery period: (time 300 min, drug: 0.9 % NaCl , 2ml/h infusion). There were no significant differences between control and ketanserin groups during basal period, on occlusion period and 120th minute of period in creatinine clearance, urine output, fractional sodium excretion (FENa) and mean arterial pressure (MAP) (p>0.05). At the 300th min of the recovery period creatinine clearance, urine output and MAP values of ketanserin group were significantly higher than control. There was a significant difference between ketanserin and other group (p<0.05). FENa values were found to be significantly lower than other group's (p<0.05). Our results verified that ketanserin, a 5-hydroxytriptamine-2 (5HT2) receptor blocker, had a positive effect on renal functions in post ischemic rat kidney. In this observation we conclude that serotonin plays a major role on postischemic vasoconstriction and 5HT2 receptor blockers diminish this effect.
