Evaluation of chitosan used as an excipient in tablet formulations

Özet

Chitosan was chosen as a model excipient because it has many different functions in pharmaceuticals. This study is intended to investigate the use and optimum concentration of chitosan as a tablet binder and disintegrant. Chitosan was prepared by two tabletting methods. In these methods, piroxicam was selected as an active substance. Tablet formulations were prepared by granulating three viscosity grades of CMC (carboxymethylcellulose) and three viscosity grades of PVP (poly-(N-vinylpyrrolidone)) in varying ratios. Chitosan was evaluated as a binder for piroxicam tablets and compared with other polymer binders such as PVP. Again, a chitosan was evaluated as a disintegrant for piroxicam tablets as compared with other cellulose disintegrants such as CMC. In the wet method, chitosan was found to decrease the release of the active substance. Therefore, chitosan is considered to be useful as excipient in the drug forms to be releasing an active substance in a controlled way. Chitosan is a pharmaceutical excipient with natural origin that may contain binding and disintegrating properties together. It is considered, that the use of chitosan in pharmaceutical excipients that are used in fewer kinds and amounts in the formulations, may increase. From the economical point of view, this situation may provide superiority.
