Intrasplenic hepatocyte transplantation in rats: A preliminary report

Background/aims: Currently available evidence suggests that hepatocyte transplantation has significant potential for a variety of clinical applications, including acute and chronic liver diseases. In addition, hepatocyte transplantation may be of use for gene directed therapies. Based on this evidence, interest in hepatocyte transplantation has been widely increasing. The aim of this study was to isolate and transplant hepatocytes into the spleens of recipient rats and to determine their fate histologically to establish basic conditions for further studies. Methods: Donor hepatocytes were isolated from rat livers by modified two-step collagenase digestion method and isolated hepatocytes were transplanted intrasplenically. All recipient rats received cyclosporin for three weeks. Spleens of recipient rats were removed for histopathological examination three weeks after hepatocyte transplantation. Results: Groups of hepatocytes showing typical morphology were detected in the spleen tissue of recipient rats, reflecting successful engraftment of transplanted hepatocytes. Conclusion: Hepatocyte transplantation is an exciting and developing field of hepatology. It is believed that more studies are needed to explore the potential role of hepatocyte transplantation in liver disease.
