Determination of antioxidant activities and total phenolic contents of grapefruit and seed extracts of Kilis Karasi {vitis vinifera l.)

In this study, the antioxidant activities of ethanol and acetone extracts of Kilis Karasi grapefruit and seeds at different concentrations (20, 40 and 80 |j.g/mL) were investigated by various in vitro antioxidant assays, including 2,2-diphenyl-l-picrylhydrazyl free radical (DPPH) scavenging and 2,2'- azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging activities and reducing power. The results were compared to the standard antioxidants such as butylated hydroxyanisole (BHA) and Vitamin C. Total phenolic content of both extracts was also determined. DPPH radical scavenging activities of ethanol extracts of Kilis Karasi grapefruit, seeds and standards were in the order of Vitamin C, Seeds, BHA and Grapefruit and found as 97.0 %, 96.5 %, 94.8 % and 18.9 % at the concentration 80 jj,g/mL, respectively. The scavenging effects of acetone extracts of Kilis Karasi grapefruit, seeds and standards on the DPPH radical scavenging activities were ordered as Vitamin C, BHA, Seeds and Grapefruit and results were presented as 94.7 %, 94.5 %, 89.4 % and 4.77 % at 80 |ig/mL concentration, respectively. ABTS radical scavenging activities of ethanol extract at 80 jj,g/mL concentration were ordered as Vitamin C, Grapefruit, BHA and Seeds and were presented as 96.7 %, 94.4 %, 93.8 % and 81.2 % respectively. ABTS radical scavenging activities of acetone extract at the same concentration were ordered as Vitamin C, BHA, Grapefruit and Seeds and results were presented as 98.1 %, 92.2 %, 40.9 % and 16.9 %, respectively. At the concentration 80 |j.g/mL for ethanol extracts reducing power was in the following order: Vitamin C, Seed, BHA and Grapefruit, and at the same concentration for acetone extracts was in the following order: Vitamin C, BHA, Grapefruit and Seed. Total phenolic content in ethanol and acetone extracts was found for Grapefruit as 21.7 and 156 mg GAE/g extract and for Seeds as 67.8 and 84.4 mg GAE/g extract, respectively. In both extracts Kilis Karasi Grapefruit and Seeds exhibited high antioxidant activities and consisted of high phenolic compounds content. Therefore, these natural products easily can be used as antioxidant agents for health.
