The effectiveness of pentoxifylline, either alone or in combination with indomethacin, in early experimental stroke

The inflammatory response plays an important role in the potentiation of central nervous system ischemic injury. In the present study we investigated whether pentoxifylline alone and in combination with indomethacin alter the serum level of interleukin-6 and tumor necrosis factor-alpha and if so, such changes cause stroke volume reduction or functional recovery on experimental stroke. The study was done in a model of permanent occlusion of middle cerebral artery using the intraluminal nylon filament suture in mice. Male Balb/C mice were divided into five groups: Pentoxifylline (30 and 100 mg/kg/dose); pentoxifylline (100 mg/kg/dose) + indomethacin (100 mg/kg/dose); placebo (0.3 ml saline) and sham (only carotid ligation + 0.3 ml saline). Interleukin-6 and tumor necrosis factor-alpha levels of placebo, 30 and 100 mg/kg pentoxifylline, pentoxifylline + indomethacin, and sham groups were 11.19, 14.08, 10.84, 10.09, 3.34 and 60.73, 67.01, 72.41, 74.37 and 6.24 pg/ml respectively. Infarct volume of placebo, pentoxifylline (30 and 100 mg/kg), and pentoxifylline + indomethacin groups were 23.11, 20.80, 20.28, and 19.76%. Post-stroke body swing asymmetry index of placebo, pentoxifylline (30 and 100 mg/kg), pentoxifylline + indomethacin, and sham groups were 13.50, 14.00, 14.44, 14.20, and 3.66 respectively. Data were analyzed by Kruskal-Wallis test and Mann-Whitney test with Bonferroni correction. Pentoxifylline, either alone or in combination with indomethacin, didn't alter the levels of interleukin-6 and tumor necrosis factor-alpha, didn't reduce the stroke volume and didn't cause functional recovery in stroke animals. © Universitätsverlag Ulm GmbH 2006.
