Resveratrol-loaded bioinspired dipeptide nanogels for anti-aging and wound healing applications

Özet

Nowadays, the increasing desire to look good has caused an increase in interest in products applied to the skin. The failure to obtain the desired efficiency from the products used for this purpose and the encounter of undesirable effects have also led to approaches with products of natural origin. Here, Fmoc-diphenylalanine (Fmoc-FF) dipeptide nanogel structures loaded with resveratrol (trans-RSV), which is known to have positive effects on the skin, were created and the effects of these structures on hydrogen peroxide (H2O2)-induced skin aging and wound were evaluated. The obtained nanogel structures were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR) and differential scanning calorimetry (DSC) and then their trans-RSV release behavior was investigated. In order to evaluate the effects on aging, an aging model was created in human keratinocyte (HaCaT) cells with H2O2, and the effects on cytotoxicity, genotoxicity, ROS release, oxidative stress and inflammatory biomarkers were evaluated. Stratch assay was used to investigate wound healing. When all the results were evaluated together, it was observed that trans-RSV-loaded Fmoc-FF nanogels were effective against aging and wounds. However, further in vitro and in vivo studies are needed for the use of the structures.
