The effect of N-acetylcystein on radiation induced lung damage in guinea pigs

N-acetylcystein (NAC), a thiol compound, which when metabolizes to cysteine, serves as a precursor of glutathione (GSH), a free radical scavenger. In this study, the effect of NAC on decreasing radiation induced lung damage in guinea pigs by its property as a GSH precursor has been analysed. In this study 30 guinea pigs of both genders, weighing 250-350 grams were used. They were separated into three groups, each group consisting of 10 animals. To the control group, only 0.1cc/10gr intraperitoneal 0.9 % NaCl was given for five days. To the second group the same solution was given again intraperitoneally (i.p.) for 5 days followed by total body irradiation. To the third group 200mg/kg i.p. NAC phospate buffered in saline was administered for 5 days followed by total body irradiation. After the drugs were administered all guinea pigs except those belonging to the control group received total body irradiation with a total dose of 485 cGy by a Co-60 machine with SAD: 80cm. technique. During irradiation the guinea pigs were separately placed in a plexiglass container which had a transmission factor of 0.988. Glutathione (GSH), Glutathione peroxidase (GSH-Px) and Glutathione-S-transferase(GST) levels were analyzed in the extracted lung tissues 24 hours after irradiation. In addition, the lungs of the guinea pigs belonging to all three groups were histopathologically evaluated. Irradiation statistically decreased the GSH, GSH-Px and GST levels in the lung tissues when compared to the control group. However in the group to which NAC was administered along with irradiation, a significant increase was seen in the levels of GSH, GST and GSH-Px enzymes when compared to the group which was solely irradiated. Histologically the group which was both irradiated and administered NAC had similar properties to the control group whereas, in the group which was irradiated only, there was a degeneration especially in type II cells. In irradiated patients N-Acetyl cystein may be helpful in preventing lung damage.
