Özet

In this study, we aimed to determine the effects of ketorolac tromethamine (KT) on renal tubular cell functions in different usage times. Forty healthy guinea pigs assigned to two groups were studied. Group I was divided into two subgroups as Group IP (0.3 ml % 0.09 NaCl IM for two days) and Group IK (KT 3 mg/kg IM in 0.3 ml volume for two days). Group II was also divided into two groups as Group IIP and Group IIK with the same % 0.9 NaCl and KT doses for seven days. Following the last injection urine was collected for 24 hours to determine NAG levels, proteinuria, hematuria, pyuria and density. Nephrectomy was performed to assess any histopathological change. Although no significant difference was observed in urine NAG levels between the placebo and KT groups; it was found significantly higher in Group IK than Group IIK (p<0.05). Pyuria, hematuria and changes in density were observed in none the groups. While no proteinuria was found in placebo groups, statistically insignificant proteinuria was determined in 20 % and 30 % of Group IK and Group IIK, respectively. In histopathological examination glomerular atrophy, crescent formation, tubular necrosis, edema and fibrosis was not observed in all the groups. In all of the cases polymorphonuclear cell infiltration, interstitial hyperemia and inflammation, tubular atrophy and calcification, with statistically insignificant difference between the groups was detected. As a result of our study we conclude that KT usage, in these doses and time intervals, causes no clinically important renal function disturbances. In view of observing tabular atrophy and calcification, particular for nonsteroidal antiinflammatory drug usage, more detailed histopathological investigations are needed.
