Protective effect of nebivolol in impairment of endothelium-dependent relaxation caused by high glucose in rat aorta

Nebivolol has endothelium-protective effects in cardiac diseases. However, experimental findings regarding protective role of nebivolol inthe hyperglycemia-caused endothelial dysfunction are still limited. This study aimed to investigate the protective effect of nebivolol against impairment of endothelium-dependent relaxation (EDR) caused by high glucose (HG). The aortas isolated from Sprague-Dawley rats were subjected to different incubations in organ baths for the following groups: control, control+nebivolol, HG, HG+nebivolol, and mannitol. Afterwards, cumulative concentration response curves (CCRCs) to acetylcholine and nebivolol were generated in precontracted aortic rings from all groups and control and HG groups, respectively. Incubation of aortic rings with HG resulted in a significant inhibition of EDR.This inhibition caused by HG was reversed to control after co-incubation with nebivolol. The relaxations in response to nebivolol were not different between aortic rings from control and HG groups.These findings demonstrate that nebivolol protects against the endothelial dysfunction caused by HG and preserves its vasorelaxant effect in aortas exposed to HG.
