Bacterial contamination in local anaesthetic ampules

Özet

We investigated the bacterial contamination in ampules of local anaesthetics. We formed 6 groups of 3 ampules of lidocaine and 3 ampules of bupivacaine in each group. The ampules in the first group were opened without applying anything at their neck. The ampules in the second group were opened following alcohol swabbing. While third group ampules were opened following povidon iodine swabbing. The ampules of the fourth, fifth and the sixth groups were first contaminated with Staphylococcus aureus; then the fourth group's ampules were opened without any preparation while the fifth group's ampules were swabbed with alcohol and the sixth group's ampules were swabbed with povidon iodine before opening. The content of the glass ampules were taken into the injector at once and placed firstly on the bouillon and then on bloody plaque. The growth of 1 colony of S. aureus in one of bupivacaine ampules in group II, and of 1 colony S. epidermidis in one of lidocaine ampules in group III were not regarded remarkable. The growing of 1,000 colonies of S. aureus in one ampule and 10,000 colonies in another ampule of group IV ampules were regarded significant (p < 0.05). Growth was not observed in bupivacaine and lidocaine ampules which were comtaminated with S. aureus and swabbed with alcohol (group V) or povidon iodine (group VI) (p < 0.05). As a result, it was concluded that swabbing the neck of local anaesthetic ampules before opening and care for the sterilization would decrease the risk of contamination.
