In vitro toxicology studies in neuroblastoma cells

Özet

Neuroblastoma (NB) is the most common solid tumor in early childhood which is derived from the peripheral sympathetic nervous system of the neural crest. Although the prognosis for low-risk NB patients is good, the 5-year survival rates for high-risk and relapsed patients are low despite aggressive multimodal therapy which is a combination of surgery, radiation therapy, and intensive chemotherapy. Therefore, there is a need to develop novel treatment strategies to control NB. Beneficial effects of some natural compounds such as betulinic acid, buchelin derivates from Lauraceae family, Aloe vera extract, Rose Bengal, quercetin, epigallocatechin- 3 - gallate, and Huaier extract have been investigated by cytoxicity and genotoxicity assays in NB cells. While comet assay was the most used technique for the evaluation of genotoxicity, MTT (3-(4,5-Dimethylthiazol-2-yl), XTT (sodium 3'-[1- (phenylaminocarbonyl)- 3,4-tetrazolium]-bis (4-methoxy6-nitro), propidium iodide, trypan blue, and alamar blue assays were commonly used to evaluate the cytotoxic effects in these studies. In addition to natural compounds, research focused on the effects of drugs, and also novel compounds on NB cells. For example, lithium, nickel, etoposide, bortezomib, sorafenib, nifurtimox synthetic retinoid RO 13-6307, and novel small molecule focal adhesion kinase inhibitor TAE 226 have shown some beneficial effects on NB. This chapter aims to provide current knowledge about in vitro toxicology studies in neuroblastoma cells especially focused on the treatment of NB.
