Özet
The consistent interaction between the cells and the extracellular matrix components of the microenvironment in the bone marrow are provided by some molecules, factors and signals. Adipocytes, the components of bone marrow stromal cells, have active roles in protein synthesis, energy regulation, inflammation, immune response and hematopoiesis. The regulatory effects of cytokines released from adipocytes on proliferation and differentiation of hematopoietic cells have been determined. Leptin is a 16 kDa, non-glycolised, small protein that is mainly synthesized in adipocytes. It is also shown to be synthesized in hypothalamus, pituitary gland, stomach, intestine, skeletal muscle, mammary epithelium, condrocytes, placenta, cartilage and bone cells, stromal cells of bone marrow and immunocytes. Serum leptin levels increase in relation with body mass index and body fat tissue. Leptin controls food intake and body temperature by influencing hypotalamus. The level of leptin in circulation reflects the amount of energy stores of adipose tissue and adipose mass ratio of body. Also, leptin is an adipokine required for normal lymphopoesis which stimulates the proliferation/differentiation of myelocytic, monocytic, erythroid and megakaryocyte precursors. Leptin is thought to be a triggering factor for hematological malignancies, because of its capacity for stimulation of hematopoetic precursor cells. Several studies have shown the relationship between leptin and hematological malignancies. Leptin may be useful in bone marrow transplantations performed by collecting peripheral stem cells for its positive efect on primitive and mature series of hematopoietic cells. Also, it may be an innovator for studies that intend to increase the amount of hematopoietic stem cells by pharmacological use of leptin. Copyright © 2012 by Türkiye Klinikleri.
