Özet

Propofol, a relatively new intravenous anaesthetic agent, has been proposed to have antinociceptive effects on the spinal cord when given intravenously. The present study was designed to examine whether intrathecally administered propofol has antinociceptive effects. 18 Chinchilla rabbits allocated in 2 groups were studied. The cisterna magna was aseptically punctured by a 22-gauge butterfly needle after anesthesia induction with ether. When the animal recovered from ether anaesthesia, Group I received 4 mL 0.2 mg/kg propofol intrathecally and Group II 4 mL 5% dextrose intrathecally. The electrical stimulies were administered in a steady increasing voltage of 1 volt. When the animal's pain threshold was reached, noticeably by leg retraction, that value was considered the nociceptive level. Measurements were done before the anaesthesia induction (baseline) and 5, 15, 30, 45, 60 minutes after intrathecal injections. Pain thresholds were higher at the 5th and 15th minutes after the propofol injection and returned to baseline values at the 30th minute. The results of our study demonstrate that intrathecally administered propofol may have antinociceptive effect. The duration of this effect was short and it may be related to propofol's extensive distribution and rapid elimination.
