It is known that polymorphonuclear leukocyte (PNL) functions and immunity are defective in patients with chronic renal failure (CRF). Granulocyte-colony stimulating factor (G-CSF) has been reported to restore the defective PNL functions. In this study, the in-vitro effect of G-CSF on the functions of PNLs obtained from CRF patients has been investigated. Fourteen patients who were on hemodialysis programme at our hospital's hemodialysis unit and 14 healthy volunteers were included into the study. PNLs taken from each group were treated by increasing concentrations of G-CSF, and PNL functions, such as phagocytosis of candida cells, intracellular killing, uptake and phagocytic index were evaluated. PNLs of CRF patients had lower functions than those of the control group. G-CSF had no significant effect on the functions of PNLs in healthy volunteers. Phagocytosis and intracellular killing increased significantly at 4500 and 6000 U/ml concentrations of the G-CSF in CRF group. It has been concluded that PNLs of the CRF patients have defective functions and G-CSF treatment may have a positive effect on PNL functions in these patients.
